Vonorazan (CAS number: 881681-00-1), chemical name 1-[5-(2-fluorophenyl)-1-[(pyridin-3-yl)sulfonyl]-1H-pyrrol-3-yl]-N-methylmethanamine, is a new type of anti-gastric acid drug, which belongs to potassium competitive acid blocker (P-CAB). Developed by Takeda Pharmaceutical in collaboration with Otsuka Pharmaceutical, Wonolazan is used to treat gastric acid-related diseases such as erosive esophagitis, gastric ulcers, and duodenal ulcers.

    The molecular formula of Vonorazan is C17H16FN3O2S and the molecular weight is 345.39. It is a proton pump inhibitor (PPI) with highly potent oral activity and is able to inhibit H+,K+-ATPase enzyme activity in porcine gastric microsomes at pH 6.5 with an IC50 value of 19 nM. Wonoprazan was developed to study and treat gastric acid-related diseases such as gastroesophageal reflux disease and peptic ulcer, and can also be used for the eradication of Helicobacter pylori.
    The physicochemical properties of Vonorazan include a predicted boiling point of 530.3±60.0 °C, a density of 1.31±0.1 g/cm3, and storage conditions requiring storage in a freezer at -20 °C and being carried out in an inert atmosphere to maintain its stability. It is soluble in chloroform, DMSO, and methanol, and is hygroscopic. The predicted acidity coefficient (pKa) of Vonorazan was 9.06±0.10, and the morphology was solid, with a pale yellow to pale yellow color.
